The high blocking efficiency of casein is due to its content of small molecular weight proteins. The solution is ready for dilution to specific application requirements.
LICOR Intercept T20 (TBS) Protein-Free Antibody Diluent (500 mL)
Intercept T20 (TBS) Protein-Free Antibody Diluent improves the specificity of the primary and secondary antibodies, reducing off-target effects. The diluent contains Intercept (TBS) Protein-Free Blocking Buffer in Tris-buffered saline (TBS) preformulated with Tween 20. This diluent can be used where animal-based products are prohibited. Since there’s no need to mix the diluent yourself, it saves you time and reduces potential variation. Intercept T20 (TBS) Protein-Free Antibody Diluent contains a 0.05% concentration of Tween 20. It contains no sodium azide and is stored at 4 °C.

The blocking buffer is to block excess binding sites on the micro-plate for reducing non-specific binding signal, by which the assay sensitivity and specificity are improved. As many factors can influence nonspecific binding, there is no single blocking buffer that is ideal for each assay. For example, depending on the recognition of detection probes from different assays, one individual blocking buffer may cross-react with detection probe. StrongBlock (I, II, III) are a variety of buffers designed by different blocker components.

Novocastra Peroxidase Block, RE7101, is intended for use in the peroxidase based immunohistochemical (IHC) staining procedures. The presence of pseudoperoxidase (erythrocytes) and endogenous peroxidase in paraffin sections to be stained by immunoperoxidase procedures, can result in nonspecific staining. A method for the blocking of pseudoperoxidase was described (StreefkerkJG, Journal of Histochemistry and Cytochemistry. 20: 829 (1972)). This product is used in a peroxidase based IHC procedure. Incubating sections with Novocastra Peroxidase Block, RE7101, can neutralize endogenous peroxidase activity. 25 mL of reagent is supplied.

It is ideal for assays that require high reproducibility and low coefficients of variation, such as assays for pre-clinical and clinical studies, production of high quality diagnostics or measurement of precious specimens. The Blocking Solution is based on highly purified casein that has been chemically modified for maximum adsorption efficiency and a dense blocking layer. By incubating a surface with The Blocking Solution, such as a microtiter plate or western blotting membrane, free spaces on the surface are saturated by the components of this blocker. This minimizes unwanted binding of the analyte or the detection antibody to the surface, avoiding background and improving sensitivity of the assay.
